About AICAR
"AICAR (5-Aminoimidazole-4-carboxamide-1-β-D-ribofuranosyl 5'-monophosphate, also known as Acadesine) is a naturally occurring purine biosynthesis intermediate conserved across all organisms. Its core mechanism of action is as a potent activator of AMP-activated protein kinase (AMPK)—it mimics cellular energy depletion without altering ATP, ADP, or AMP levels, thereby activating AMPK and suppressing anabolic processes. At the cellular and animal level, AICAR promotes non-insulin-dependent glucose uptake in skeletal muscle via AMPK activation, a pathway not blocked by PI3K inhibitors.
AICAR received Orphan Drug designation in the European Union for the treatment of adult B-cell chronic lymphocytic leukemia. Currently, the compound is primarily used as a research tool and experimental agent."
Main functions
"1.Improves Metabolic Syndrome and Type 2 Diabetes: In animal models, AICAR reduces body weight and visceral fat, improves glucose and insulin levels, and alleviates high-fat diet-induced metabolic disorders. Systemic administration in humans reduces hepatic glucose production and increases skeletal muscle glucose uptake.
2.Anti-inflammatory and Pro-resolving Properties: By inducing macrophage polarization toward a pro-resolving M2 phenotype, AICAR increases anti-inflammatory IL-10 and reduces pro-inflammatory IL-1β, alleviating inflammatory edema and exerting analgesic effects. In acute lung injury models, AICAR provides protection via the AMPK/Nrf2/HO-1 pathway.
3.Antiproliferative and Anticancer Activity: AICAR demonstrates cell growth inhibition and apoptosis induction in various tumor cell lines and xenograft models. Its antiproliferative effects are linked to disruption of the AICAR/NTP balance, with selective induction of apoptosis in aneuploid cells.
4.Exercise Mimetic Effects: AICAR recapitulates certain metabolic adaptations of exercise training in sedentary mice, activating AMPK and increasing skeletal muscle glucose uptake and fatty acid oxidation.
5.Anti-ischemic and Cardioprotective Effects: Clinical studies show that AICAR reduces the duration of myocardial ischemia following coronary artery bypass surgery. In cardiomyocytes, AICAR induces metabolic remodeling that redirects free fatty acids toward neutral storage, potentially improving ischemia-reperfusion injury in diabetic patients."
