About Gonadorelin
"Gonadorelin is a synthetic decapeptide (sequence: pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH₂) that is chemically and physiologically identical to endogenous human gonadotropin-releasing hormone (GnRH). Its core mechanism of action is to stimulate the anterior pituitary to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which in turn regulate gonadal steroid production.
The biological effect of gonadorelin is biphasic and depends on the mode of administration:
Pulsatile Administration: Mimics physiological hypothalamic pulsatile secretion, transiently stimulating LH and FSH release—used for diagnosis and treatment of hypogonadism.
Continuous Administration: Prolonged high-level exposure causes pituitary receptor down-regulation, inhibiting gonadotropin secretion—this forms the basis for using GnRH analogues (e.g., leuprorelin, goserelin) in hormone-dependent cancers.
Native gonadorelin has an extremely short plasma half-life (only minutes), limiting its clinical utility and typically requiring administration via infusion pumps."
