Product profile

About KPV

KPV is a synthetic tripeptide composed of lysine (K), proline (P), and valine (V), corresponding to the C-terminal sequence (amino acids 11–13) of α-melanocyte-stimulating hormone (α-MSH). As an endogenous peptide, KPV retains the core anti-inflammatory activity of α-MSH while completely eliminating its pigmentary (melanogenic) effects, making it a potentially safer anti-inflammatory candidate.

Its mechanism of action is distinct. Research confirms that KPV does not exert its effects primarily through classical melanocortin receptors but rather through the inhibition of the NF-κB signaling pathway. Specifically, KPV suppresses nuclear translocation of p65RelA and stabilizes IκBα to block the inflammatory cascade. Furthermore, KPV can be actively transported into cells via the PepT1 transporter expressed on intestinal epithelial and immune cells, enabling it to exert anti-inflammatory effects in tissues such as the gut. Recent studies have also highlighted KPV's potent antioxidant activity, as it scavenges reactive oxygen species (ROS) and inhibits MAPK/NF-κB pathways and pyroptosis, protecting cells from damage induced by environmental pollutants such as fine particulate matter (PM10).

Main functions

  • 1.Potent Anti-Inflammatory and Immunomodulatory Effects: Inhibits NF-κB activation and reduces pro-inflammatory cytokines (e.g., IL-1β, IL-8), demonstrating efficacy across multiple inflammatory models.
  • 2.Skin Protection and Anti-Aging: Protects keratinocytes from oxidative damage and inflammation induced by environmental pollutants (PM10) by scavenging ROS and inhibiting pyroptotic signaling pathways, showing potential for functional cosmetics and dermatological treatments.
  • 3.Supports Gut Health: In animal models, oral KPV alleviates symptoms of DSS- and TNBS-induced colitis, including weight recovery and reduced histological inflammation, suggesting it could be a novel therapeutic option for inflammatory bowel disease (IBD).
  • 4.Potential Cardiovascular Protection: A recent study demonstrated that carrier-free nanoparticles self-assembled from KPV and rapamycin effectively inhibit vascular calcification in mice via anti-inflammatory and autophagy-activating mechanisms.
  • 5.Unique Mechanism of Action: Unlike many peptides that act via GPCRs, KPV primarily functions through direct modulation of intracellular transcription factors (NF-κB) and transport proteins (PepT1), offering a distinct mode of action and potential for synergistic applications.

Recommended audience

  • Individuals focused on managing chronic inflammation (e.g., autoimmune-related inflammation, irritable bowel syndrome).
  • Health-conscious individuals seeking to improve skin health and combat environmental damage and photoaging.
  • Individuals seeking gut health support and adjunctive management for inflammatory bowel conditions.
