About Melanotan I
"Melanotan-1 (MT-1, also known as Afamelanotide) is a synthetic tridecapeptide (sequence: Ac-Ser-Tyr-Ser-Nle-Glu-His-D-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH₂) and a super-potent analogue of α-melanocyte-stimulating hormone (α-MSH). Its key structural modifications—substitution of methionine with norleucine (Nle) at position 4 and introduction of D-phenylalanine (D-Phe) at position 7—confer significantly enhanced receptor affinity and biological activity compared to native α-MSH, while also improving metabolic stability.
MT-1 acts as a selective agonist of the melanocortin-1 receptor (MC1R) on skin melanocytes, stimulating tyrosinase activity and promoting melanin synthesis and deposition, thereby inducing skin pigmentation (tanning) that can occur even without ultraviolet exposure. The compound completed Phase I clinical trials in 2004, and its successor analog, afamelanotide, has since been approved in the EU and US for specific therapeutic indications."

"1.Synergistically Enhances UV-Induced Tanning: Phase I clinical studies demonstrated that MT-1 combined with UV-B light or sunlight produces a marked synergistic tanning effect, with significantly greater skin darkening than UV light or drug alone. The tanning effect was maintained at least 3 weeks longer than in sunlight-only controls, who required 50% more sun exposure for equivalent tanning.
2.Reduces UV-Induced Skin Damage: In the 0.08 mg/kg/day cohort, MT-1-treated subjects exhibited 47% fewer sunburn cells at irradiated sites, suggesting photoprotective effects and potential for prevention of sunlight-induced skin cancers.
3.Reduces UV Exposure Requirements: By significantly enhancing photosensitivity, MT-1 allows users to achieve desired pigmentation with substantially less cumulative UV exposure.
4.Favorable Safety Profile: Phase I trials reported no pathological findings, with adverse effects limited to mild nausea and transient facial flushing. Daily dosing at 0.16 mg/kg for 20 days over 4 weeks produced no cumulative or novel adverse effects."
