Product profile

About Melatonin

"Melatonin (N-acetyl-5-methoxytryptamine) is a neuroendocrine hormone primarily secreted by the pineal gland, often referred to as the ""hormone of darkness."" Its secretion is regulated by the suprachiasmatic nucleus (SCN) of the hypothalamus and follows a circadian rhythm—low during the day and high at night—with darkness stimulating secretion and light inhibiting it. Melatonin is also synthesized in other sites, including the retina, bone marrow, and gastrointestinal tract.

Its primary function is circadian rhythm synchronization: acting on high-affinity MT1 and MT2 receptors in the SCN to regulate sleep-wake cycles, body temperature, hormone secretion, and other physiological rhythms. MT1 receptors primarily suppress SCN neuronal firing to promote sleep onset, while MT2 receptors mainly mediate circadian phase shifts. Melatonin also directly scavenges free radicals (receptor-independently), providing antioxidant and anti-inflammatory effects."

Main functions

  • "1.Regulates Circadian Rhythms and Improves Sleep: Melatonin acts as a ""darkness signal"" to synchronize the biological clock, helping to shorten sleep onset latency and improve sleep quality in conditions like jet lag, shift work disorder, and certain types of insomnia.
  • 2.Antioxidant and Cytoprotective Effects: As a direct free radical scavenger, melatonin neutralizes harmful reactive oxygen and nitrogen species (e.g., hydroxyl radicals, peroxynitrite) and stimulates antioxidant enzymes to protect cells from oxidative damage.
  • 3.Investigational Advanced Applications:
  • Cardioprotection: In a 2025 study, researchers designed heart-targeting melatonin self-assembling peptides (Mel-NMs). In a mouse myocardial ischemia-reperfusion model, this nanodrug significantly improved cardiac function, reduced cardiomyocyte apoptosis, and lowered oxidative stress levels, showing superior cardioprotective effects over free melatonin.
  • Biological Rhythm Regulation and Anti-Aging: In preclinical studies, combining melatonin with pineal peptide preparations (e.g., Epithalamin/Epitalon) restored circadian rhythms of melatonin and cortisol production in aged rhesus monkeys. Clinical trials have also shown retinal function improvement and geroprotective potential.
  • Central Nervous System: Research indicates melatonin may produce analgesic effects through the endogenous opioid system. One study synthesized a chimeric peptide combining melatonin and deltorphin II (Del II-MT), acting as a dual receptor agonist co-activating opioid and melatonin receptor systems to produce significant, long-lasting analgesia in mouse models."

Recommended audience

  • "General Population: Individuals with circadian rhythm disturbances (e.g., jet lag, shift work), sleep onset difficulties, or poor sleep quality.
  • Specific Patients: As an adjunctive supplement in certain neurodegenerative, cardiovascular, or cancer-related conditions (under medical guidance).
  • Researchers: Scientists conducting studies on peptide-melatonin combinations, cardioprotective nanodrug development, or circadian rhythm and aging mechanisms."
