About PE-22-28
PE 22-28 is a synthetic heptapeptide (sequence: GVSWGLR) developed through structural optimization of the endogenous peptide spadin (PE 12-28). Spadin itself possesses antidepressant activity by specifically inhibiting the TREK-1 potassium channel, but it suffers from short duration of action and poor stability in vivo. By analyzing the degradation products of spadin in blood, researchers designed the shorter PE 22-28. Compared to spadin, PE 22-28 exhibits significantly higher affinity and inhibitory activity against the TREK-1 channel, with an IC50 value as low as 0.12 nM, compared to spadin's IC50 of approximately 40-60 nM—a difference of several hundredfold.

2.Rapid Antidepressant Effects: In animal behavioral models (e.g., forced swimming tests), PE 22-28 exhibits significant antidepressant activity, with observable effects after just 4 days of subchronic treatment—superior to the 3-4 week onset required by conventional antidepressants.
3.Induces Neurogenesis and Synaptogenesis: After only 4 days of treatment, PE 22-28 promotes neurogenesis in the mouse hippocampus and enhances synaptogenesis in cortical neurons (as evidenced by increased PSD-95 expression).
4.Prolonged Duration of Action: The duration of action following a single administration of PE 22-28 is extended from spadin's 7 hours to 23 hours.
