PEG-MGF is a polyethylene glycol (PEG)-modified analogue of Mechano Growth Factor (MGF). MGF is an alternatively spliced variant of the insulin-like growth factor-1 (IGF-1) gene that is locally expressed in skeletal muscle in response to mechanical load, such as resistance training. Native MGF has an extremely short half-life (only a few minutes). PEGylation—the covalent attachment of PEG molecules to the MGF molecule—significantly extends its half-life, allowing it to remain in circulation at more stable concentrations for a longer duration while reducing renal clearance and immunogenicity. PEG-MGF's mechanism of action involves activating IGF-1 receptors (IGF-1R) to promote the proliferation and differentiation of muscle stem cells (satellite cells), increasing myonuclear number and laying the cellular foundation for muscle fiber growth and repair. Its unique role lies in MGF's primary action on satellite cell activation and proliferation, rather than directly promoting protein synthesis, making it functionally complementary to factors such as IGF-1. It is typically administered via immediate post-exercise local (intramuscular) injection, 2-3 times per week.
