About Semax
Semax is a synthetic heptapeptide (sequence: Met-Glu-His-Phe-Pro-Gly-Pro) and a synthetic analogue of the adrenocorticotropic hormone (ACTH) fragment 4-10. The addition of a C-terminal Pro-Gly-Pro (PGP) tripeptide significantly enhances its metabolic stability and resistance to enzymatic degradation. The drug is approved for medical use in Russia and is included on the Russian List of Vital & Essential Drugs, primarily for the treatment of stroke, transient ischemic attacks, and cognitive disorders. It can cross the blood-brain barrier, and has gained international attention as a research compound with purported neuroprotective and nootropic properties.
Main functions
"1.Cognitive Enhancement: Semax functions as a nootropic agent, facilitating learning and memory formation. In rat models of chronic cerebral ischemia, intranasal administration has been shown to improve learning processes and both short- and long-term memory. The mechanism is linked to the modulation of neurotrophin expression, such as BDNF, in the hippocampus.
2.Significant Neuroprotection: In ischemia models, Semax activates neurotrophic factors and their receptors, promotes neuronal survival, and improves cerebral circulation, providing neuroprotective and restorative effects.
3.Anti-Amyloidogenic Potential: Recent in vitro studies demonstrate that Semax effectively inhibits the fibrillogenesis of beta-amyloid protein (Aβ), especially in the presence of copper ions, suggesting promising potential in research into neurodegenerative diseases like Alzheimer’s.
4.Unique Metal Ion Chelation: Research shows that Semax forms a very stable complex with copper ions (Cu²⁺) in an albumin-like mode. This chelating activity may help explain its ability to counteract copper-induced cytotoxicity and modulate metal-related pathologies."
