SS-31 (also known as Elamipretide, MTP-131, or Bendavia) is a synthetic tetrapeptide (sequence: H-D-Arg-Dmt-Lys-Phe-NH₂) belonging to the Szeto-Schiller (SS) peptide family. It is uniquely characterized by its ability to selectively target the inner mitochondrial membrane through high-affinity binding to cardiolipin, achieving over 1,000-fold mitochondrial enrichment independent of membrane potential. This precise targeting mechanism stabilizes mitochondrial membrane structure, reduces reactive oxygen species (ROS) production, enhances ATP synthesis, and inhibits mitochondrial permeability transition pore (mPTP) opening. Based on its broad mitochondrial protective effects, SS-31 has demonstrated therapeutic potential in various disease models associated with mitochondrial dysfunction. Notably, SS-31 (Elamipretide) has recently received accelerated FDA approval as the first treatment for Barth syndrome.
Main functions
