About Thymosin Alpha-1
Thymosin alpha-1 (INN: thymalfasin) is a synthetic 28-amino acid peptide and the first peptide isolated from thymic tissue with demonstrated immunomodulatory activity. Its amino acid sequence is highly conserved across bovine, porcine, ovine, and human species, suggesting significant biological importance. As a biological response modifier, thymosin alpha-1 acts through Toll-like receptors (TLR-9 and TLR-2) on myeloid and plasmacytoid dendritic cells, promoting the production of immune-related cytokines and enhancing immune responses. The drug is approved in over 35 countries worldwide for the treatment of chronic hepatitis B and hepatitis C, marketed under the brand name Zadaxin®.
Main functions
"1.Immunomodulation and Enhancement: Promotes T-cell and dendritic cell proliferation, enhances antibody responses, modulates cytokine and chemokine production, and exerts bidirectional immunoregulatory effects.
2.Antiviral Applications: Approved in over 35 countries for the treatment of chronic hepatitis B and hepatitis C.
3.Oncology Adjunct Therapy: Granted FDA Orphan Drug designation for the treatment of stage IIb through stage IV malignant melanoma. A Phase II study demonstrated that adding thymosin alpha-1 (3.2 mg) to DTIC chemotherapy significantly improved overall tumor response rates from 3.9% to 12.9%.
4.Potential Anti-Infective Applications: Preclinical and clinical studies have explored its potential in septic shock, acute respiratory distress syndrome, peritonitis, cytomegalovirus infection, tuberculosis, and severe lung infections.
5.Vaccine Adjuvant: An ongoing Phase I clinical trial (NCT06821100) is evaluating the safety and efficacy of thymosin alpha-1 as an enhancer of COVID-19 vaccine response in older adults."
