About Botulinum Toxin

Botulinum toxin (BoNT) is a potent neurotoxin produced by the bacterium Clostridium botulinum and is one of the most toxic biologically active substances known. Its molecular structure consists of a heavy chain (HC) and a light chain (LC) linked by a disulfide bond. The heavy chain binds to synaptic vesicle proteins (such as synaptotagmins SYT1/SYT2) and gangliosides at the nerve terminal, mediating endocytosis and transmembrane translocation of the toxin. The light chain is a zinc-dependent endopeptidase that, upon entry into the cytosol, specifically cleaves synaptic vesicle-associated membrane proteins (e.g., VAMP2/synaptobrevin-2), preventing synaptic vesicle fusion with the cytoplasmic membrane and thereby inhibiting neurotransmitter (acetylcholine) release. This precise neural blockade forms the core molecular basis for the therapeutic and aesthetic applications of botulinum toxin. The clinically used form is primarily type A (BTX-A), which is highly purified and employed for various medical and aesthetic indications.
